What is orforglipron?

Orforglipron is the international non-proprietary name of a small-molecule (non-peptide) agonist of the glucagon-like peptide-1 (GLP-1) receptor. Unlike injectable GLP-1 medicines, it is a tablet, and unlike oral semaglutide it can be taken at any time of day with or without food or water restrictions.[1][2]

According to the current US prescribing information (DailyMed, revised July 2026), Foundayo is indicated, together with a reduced-calorie diet and increased physical activity, to reduce excess body weight and maintain weight reduction long term in:[1]

  • adults with obesity, or
  • adults with overweight who have at least one weight-related condition.

In the United Kingdom, the MHRA authorised orforglipron on 10 August 2026 for weight management in adults and for adults with insufficiently controlled type 2 diabetes. Approved uses differ between countries.[3]

Foundayo is the originator orforglipron medicine. It was discovered by Chugai Pharmaceutical, developed by Eli Lilly and Company, and approved by the US Food and Drug Administration on 1 April 2026 as the first GLP-1 pill for weight loss that can be taken at any time of day without food or water restrictions. Other products represented as containing orforglipron are made by other manufacturers and sold under their own brand names.[1][2]

Boxed warning: risk of thyroid C-cell tumours. In rodents, GLP-1 receptor agonists caused thyroid C-cell tumours. It is unknown whether orforglipron causes such tumours, including medullary thyroid carcinoma (MTC), in humans. Orforglipron must not be used by people with a personal or family history of MTC or with multiple endocrine neoplasia syndrome type 2 (MEN 2).[1]

Interactions and pregnancy

  • With strong CYP3A4 inhibitors the maximum dose is 9 mg once daily; strong CYP3A4 inducers should be avoided.
  • Do not exceed 20 mg simvastatin daily.
  • Orforglipron slows stomach emptying and may change the absorption of other oral medicines.
  • People using oral contraceptives should switch to a non-oral method, or add a barrier method, for 30 days after starting and after each dose increase.
  • Orforglipron may cause harm to an unborn baby; treatment should be stopped when pregnancy is recognised.
Dosing

How orforglipron is taken (US label)

Orforglipron is taken by mouth once a day, at any time of day, with or without food. Tablets are swallowed whole and must not be broken, crushed or chewed. The dose is increased gradually to reduce stomach and bowel side effects. Your doctor decides the dose.[1]

What the main studies showed

In a 72-week study of 3,127 adults with obesity or overweight without diabetes, average body-weight change was −7.4% (5.5 mg), −8.3% (9 mg) and −11.1% (17.2 mg), compared with −2.1% with placebo. 71.5% of people on 17.2 mg lost at least 5% of their weight (placebo 26.8%).[1]

In a 72-week study of 1,613 adults with overweight and type 2 diabetes, average weight change was −5.1%, −7.0% and −9.6% for the same doses, compared with −2.5% with placebo.[1]
